研究興趣
本研究室主要的研究工作為分子酶學及蛋白質結構與功能之探討,其範圍涵蓋生物化學、生物有機、生物物理,蛋白質工程、分子生物學及細胞生物學之範疇,更拜電腦科技及網路發展一日千里之賜,涉獵結構生物學、生物資訊學及生物醫學之領域。

研究主題
1.人類粒腺體NAD(P) +-依賴型蘋果酸酶: 結構,功能及致癌機轉之探討:本研究主題是以人類粒腺體NAD(P)+-依賴型蘋果酸酶作為題材,此酶有別於另兩類異構酶,為一異位酶,以NAD+或NADP+作為輔因子,但以NADH為生理狀況下之產物,因此被認為應與能量代謝有關。此酶被認為在一些快速生長的組織中 (脾臟,胸腺,小腸黏膜等),扮演能量提供之重要角色。特別的是,癌細胞中皆有粒腺體NAD(P)+-依賴型蘋果酸酶存在,而在大部分正常細胞中則無。因此,此酶之存在被認為與細胞不正常增生及癌化有關。本人研究之重點在於蛋白質分子階層之結構與功能關係研究,所採用之方法包括定位突變,分子模擬,及X光結晶繞射等。在酶化學、分子生物學技術及蛋白質分子結構電腦模擬方面為本人之主要工作。蛋白質科學為傳統生化學之研究主題,近年來由於立體結構工具的進展而獲致重大突破。但任何結構工作最後皆需以功能及生物意義為依歸。因此本研究計畫重點在於人類粒腺體NAD(P)+-依賴型蘋果酸酶結構及功能方面之整合,從巨分子結構的角度,來瞭解酶分子所持具之驚人催化功能,進而解決生物學的複雜問題,並提供此酶在催化機制,抑制型態、結構及功能等之重要資訊來作為新藥設計的方向,最後與臨床醫學結合,發展治療癌症疾病之新方法。
2.人類第四型胜肽精胺酸脫亞胺酶功能及調控之分子機制:本研究主題是以人類四型胜肽精胺酸脫亞胺酶作為題材。此酶為催化鈣離子依賴型之脫亞胺反應,為轉變蛋白質上的精胺酸(arginine)成為瓜胺酸(citrulline)之後轉譯修飾作用。許多證據顯示PAD4與類風濕性關節炎有相關性。在本研究中,我們探討人類PAD4功能與調控的分子機制,重點在於探討此酶在鈣離子及四級結構調節之分子機制。本研究所探討的包括鈣離子的催化及調節結合位置,及探究此酵素受鈣離子調控的詳細機制、探討四級結構交互作用與此酶的酵素催化、功能及調控之關係,以及討論穩定PAD4酵素結構之因素。定點突變、酵素動力學分析和生物物理學的方法將被用來探究PAD4之催化功能和穩定性。
3.探討人類鳥胺酸脫羧酶 (ornithine decarboxylase),抗酶 (antizyme),與抗酶抑制劑 (antizyme inhibitor) 三者之間交互作用:人類鳥胺酸脫羧酶和許多種類之惡性腫瘤有著高度相關性。抗酶 (antizyme) 可直接與鳥胺酸脫羧酶結合,抑制其活性,此外,過度產生的抗酶也和抑制許多種類的癌細胞生長有關。因此,抗酶也許具有抑制腫瘤形成的功能。抗酶抑制蛋白 (antizyme inhibitor) 可使所有抗酶家族的成員失去活性,進而使鳥胺酸脫羧酶回復其活性且不受降解。由上述,推測在腫瘤形成的過程中,抗酶是負向的調控因子,而抗酶抑制蛋白是正向的調控因子。在此研究中,我們釐清了鳥胺酸脫羧酶和抗酶抑制蛋白間,其酵素活性上決定差異性之決定因子。我們將繼續研究人類鳥胺酸脫羧酶,人類抗酶抑制蛋白及錐蟲鳥胺酸脫羧酶,對於抗酶具有不同結合能力之決定因素、尋找抗酶的二級結構片段,可以有效抑制鳥胺酸脫羧酶酵素活性或是阻斷此酶形成二聚體、以及希望能找出鳥胺酸脫羧酶受質專一性之決定因子。這些結果將可應用在研發鳥胺酸脫羧酶之抑制劑而用於癌症治療上。
